ABSTRACT
Non homologous end-joining (NHEJ) is an essential DNA double-strand break (DSB) repair pathway required to maintain genome stability. We are using model unicellular organism systems, combined with biochemical and structural approaches, to understand both the mechanism and regulation of the NHEJ pathway. The simplicity of these DSB repair complexes makes them ideal models for studying both the cellular and molecular mechanisms that co-ordinate the processing and joining of DSBs by the NHEJ pathway and also provides a conceptual framework for understanding related break repair processes in higher eukaryotes.
