Abstract:

Genetic and functional characterization of lymphoid development pathways and immune responses have shown a close link with leukemia and lymphoma mechanisms and provide important insight in the role of loss of immune surveillance in tumor progression. In this context, molecular characterization of leukemia-specific enhancer deregulation and of microRNA control of B-cell differentiation and function inform us of basic mechanisms central for lymphoid development in the context of hematologic malignancies and autoimmune disease. Moving forward, as a central question in immuno-oncology, it will be essential to understand the mechanisms that control the activity of anti-tumoral and host-directed autoimmune responses upon immune-checkpoint inhibitor treatment to improve the efficacy and safety of these immunotherapies.
