Abstract:

Unravelling the mechanisms governing interactions between chemicals and DNA, our research explores DNA adducts, their interactions with repair proteins, and cellular responses, providing a roadmap for deciphering mutational signatures in cancer, biomarkers of aging and cancer drug action. Our talk synthesizes diverse strategies, with a primary emphasis on chemical-specific mapping of genome-wide chemical modifications, offering new insights into the impact of oxidation and alkylation products. Within this framework, we delve into several examples, including the genomic aftermath of exposing glioblastoma cells to the cancer drug temozolomide treatment, employing O6-MeG-seq to unravel the dynamics of O6-methylguanine formation and repair. Simultaneously, our investigation into a different cancer drug, trabectedin reveals its unique toxicity mechanism by disrupting transcription-coupled nucleotide excision repair, offering a targeted approach for precision oncology. Furthermore, the quantification and mapping of alkylation in the human genome provides insights into how environmental carcinogens shape mutational landscapes, linking in vitro damage maps to mutational signatures in human cancers. Collectively, this research leverages genome-wide profiling in order to provide new insight into early chemical interactions that drive cellular responses and disease phenotypes.
