Abstract:

Somatic mutations occur during cell division across all somatic lineages due to unavoidable DNA replication errors or damage. These mutations, also known as somatic mosaicism, are implicated in both cancer evolution and non-neoplastic disorders. Neural stem cells (NSCs) and their progenitors, which continue dividing throughout life, accumulate somatic mutations in the brain during development and aging. Over the past 15 years, our group has opened a new window into this field and provided molecular genetic evidence that somatic mutations in NSCs or their progenitors contribute to neurodevelopmental disorders and brain tumor evolution. In this seminar, I will present our recent findings on the cell of origin harboring the initial driver mutation in glioma, as well as the associated molecular evolution of these cells of origin and their tumor microenvironment.
