Abstract:

Structure-function analysis of protein complexes are crucial to understand the mode of action of the cellular machinery. In vitro techniques such as X-ray crystallography and cryo-Electron Microscopy, are fundamental to resolve the structure of proteins at high resolution. However, these approaches are limited to exploring complexes in their physiological environment. In this talk, we will discuss the strategies that we are developing to investigate the spatial arrangement of protein assemblies directly in the cellular milieu. By combining genetically encoded nanotools, light microscopy and integrative modelling, we narrow the gap between resolution and biological significance, allowing high-resolution observations in a near-physiological context. Our approach opens new avenues to investigate molecular mechanisms, including the structural dynamics that mediate the activity of the cellular machinery.
