Abstract:

Over the last decade we have decoded the influence of mutation at different scales in cancer evolution, tissue cellular population dynamics and the role in drug resistance using combined genomics and computational methods. Recently using novel single cell genome sequencing methods (Laks et al, Cell 2019) we have identified the causes and consequences of copy number structural mutations in solid cancers (Funnell et al Nature 2022), identifying genotypic resistance to DNA targeting agents (Salehi et al, Nature 2021). I will describe a novel approach to targeting cancer: we have identified small molecules that bind G-quadruplex secondary structure in the genome, with clinical activity in DNA repair deficient cancers.
