This presentation will use a biomarker discovery effort that demonstrates how a metabolomic approach can be integrated into a completely biological context. This study was successful because the results of a simple metabolomic analysis were enabled the identification of a novel biological mechanism that was verified experimentally. However, the study was fortunate in that the biomarker did not require accurate quantitation because the markers involved were either on or off as they were control features for the biological response. This is not always the case, sometimes in metabolomics the metabolic response of an organism or cell to a given stimulus is more subtle and complex. In these situations a newer technique based on isotopic internal standards, called the IROA TruQuant protocol, is required. This technique is capable of producing almost clinically accurate measurements for large portions of biochemical processes. These measurements should afford even greater understanding of the biological response, and allow one to find biomarkers for any response that are both subtle and complex.
