Abstract:
The vast majority of tissues analysed harbour stem cell populations necessary for both tissue homeostasis and repair after cellular damage. Stem cell populations, found generally in microenvironments or ‘niches’ that regulate their proliferation and differentiation, adapt their behaviour to local and systemic signals such as cell-cell signalling or ageing. One such stem-cell niche is present in the Drosophila ovary, which contains populations of germline stem cells (GSCs). Our laboratory studies this stem cell niche with a myriad of experimental approaches. Recently, we have focused our efforts on the role of niche ageing in ovary homeostasis and have described the effect of ageing on gene transcription and splicing in niche support cells.
Oogenesis is the only process in which genetic reshuffling takes place in Drosophila melanogaster, as males do not undergo recombination. Since recombination requires the generation of double strand breaks (DSBs), DNA repair mechanisms are in place in the ovary to ensure DSB damage restoration. I will present our recent results describing the regulation of DNA repair in the female germline by the transcriptional repressor Bruno 1 and how bru1 is a key factor in the mitosis-to-meiosis transition in germline cells.
