Abstract

Mammalian tissues are often composed of repeating anatomical units that are polarized by directional blood flow or morphogens. Therefore, the spatial locations of cells in tissues are critical determinants of their molecular identities. Our lab is developing approaches based on single molecule transcript imaging, laser capture microdissection and single cell RNA sequencing to obtain the global transcriptional profiles of thousands of cells while inferring their tissue locations. This enables building spatial maps of gene expression in complex organs such as the liver and the gut. I will demonstrate how these approaches uncover patterns of spatial division of labor of hepatocytes and enterocytes which facilitate optimality in these organs.
