I will describe how oncogenic fusion diversity and endogenous mutational processes shape tumour evolution and therapy response. Using CRISPR/Cas9 somatic genome editing, we generated mouse and organoid models of the major EML4-ALK variants in lung cancer. These studies reveal striking variant-specific differences in oncogenic potential, tumour suppressor interactions, and sensitivity to ALK inhibition.
In parallel, I will present data on APOBEC3B (A3B) in HER2-driven breast cancer, showing that A3B acts as both a mutagenic and selective force. High A3B expression delays tumour initiation through early DNA-damage–induced bottlenecks, yet ultimately drives chromosomal instability and aggressive evolution.
